Well since it's a "shadow" rune, it should probably act like a shadow passing through everything.
So maybe a 5% defense pierce, then 10 and 15 (of course this would be like poison damage ..... so it should probably only de-buff the enemies defense upon being hit instead of completely ignoring the enemies defense by a percentage.)
But that's my take on a "shadow" rune.
